Static blood (Oketsu) is a blood disorder of Kampo medicine. Many natural medicines have been used to cure static blood, and these drugs are called blood stasis-resolving formula. Some of these were reported to inhibit blood coagulation. Jidabokuippo (JDI), one of the blood stasis-resolving formulas, has been used to treat bruises and sprains. In this study, we evaluated the antithrombin activity of JDI and its active compounds using thrombin and an artificial substrate. Bioassay-guided isolation was performed for JDI, and the active compounds were identified using spectral data (NMR spectroscopy and mass spectrometry) by comparison with reported data. JDI depressed thrombin activity in a dose-dependent manner, ranging from 0.125 to 1 mg/mL. Chrysophanol-1- O-β-D-glucoside (1) and chrysophanol-8- O-β-D-glucoside (2) were obtained from JDI and showed thrombin inhibitory activity. This study provided the first report of the antithrombin effect of JDI and the isolation from it of two active compounds (1 and 2). Desferrioxamine B (desferal®) is the only therapeutic agent for chronic iron overload and acute iron intoxication. Therefore, new drug candidates are required for more effective treatments. Recent genome analyses of microorganisms have enabled us to identify cryptic gene clusters of secondary metabolism. In a genomic analysis of Streptomyces avermitilis, we found the putative biosynthetic gene cluster for nocardamin and desferrioxamine derivatives, the production of which by S. avermitilis had been unknown. To determine the synthesis of nocardamin and discover new derivatives in S. avermitilis, we performed a comprehensive analysis and isolation of secondary metabolites of S. avermitilis. We obtained nocardamin and its related compounds, including a derivative that initially was isolated as a microbial product. The biosynthetic pathway of nocardamin and the substrate specificity of its biosynthesis enzymes were proposed based on the structures of the isolated nocardamin derivatives. The production of nocardamin by S. avermitilis was completely suppressed by the addition of more than 5 μM ferric ions. An iron-dependent regulatory protein (IdeR)-binding motif was located upstream of the sidABCD operon, using the profile hidden Markov model method. To determine the regulation of nocardamin synthesis by IdeR in S. avermitilis, we constructed ideR deletion mutants. Desferrioxamine B (desferal®) is the only therapeutic agent for chronic iron overload and acute iron intoxication. Therefore, new drug candidates are required for more effective treatments. Recent genome analyses of microorganisms have enabled us to identify cryptic gene clusters of secondary metabolism. In a genomic analysis of Streptomyces avermitilis, we found the putative biosynthetic gene cluster for nocardamin and desferrioxamine derivatives, the production of which by S. avermitilis had been unknown. To determine the synthesis of nocardamin and discover new derivatives in S. avermitilis, we performed a comprehensive analysis and isolation of secondary metabolites of S. avermitilis. We obtained nocardamin and its related compounds, including a derivative that initially was isolated as a microbial product. The biosynthetic pathway of nocardamin and the substrate specificity of its biosynthesis enzymes were proposed based on the structures of the isolated nocardamin derivatives. The production of nocardamin by S. avermitilis was completely suppressed by the addition of more than 5 μM ferric ions. An iron-dependent regulatory protein (IdeR)-binding motif was located upstream of the sidABCD operon, using the profile hidden Markov model method. To determine the regulation of nocardamin synthesis by IdeR in S. avermitilis, we constructed ideR deletion mutants. Clinical early events in inflammation, the allergic response, and tumor metastasis involve interactions between leukocytes and endothelial cells. A variety of cytokinins and related chemical mediators control both leukocyte adhesion and subsequent intercellular invasion via the regulation and expression of cellular adhesion molecules, inhibition of cell-cell adhesion thus holds promise for the treatment of diverse pathologies. In the course of a screening program aimed at cell adhesion inhibitors obtained from microoragnisms, macrosplelides A (1) to D (4) were discovered from the fermentation broth of Microsphaeropsis sp. FO-5050. Macrosphelides are the first 16-membered-ring antibiotics embodying three lactone linkages (e.g., Macrotriolides). The macrosphelide A (1) strongly inhibit the adhesion of human-leukemia (HL-60) cells to human-umbilical-vein endothelical cells (HUVEC) in dose-dependent fasion (IC_<50>=3.5μM). Preliminary studies suggest that 1 prevents cell-cell adhesion by inhibiting the binding of sialyl Lewis^x to E-selectin. 1 also proved to be orally active against B-16 tumor metastasis to the lung in mice. We detemined the complete relative and absolute stereochemistries of 1, via X-ray crystallography and the Kakisawa-Kashman modification of Mosher's NMR method. The stereostructure of 2 was determined by chemical correlation with 1. Moreover, we accomplished the first total synthesis of 1. As our point of departure we selected the asymmetric dihydroxylation of (E,E)-hexa-2,4-dienoic acid t-butyl ester (5), which affords the (4S,5S)-diol (6) (85%ee). Selective monosilylation of 6 (TBDMSCl, DMAP, CH_2Cl_2) provided the desired ether (7). Mitsunobu inversion (PPH_3, DEAD, HCOOH; NH_3/MeOH) at C(4) of 7 furnished 8. Protection of 8 as the MEM ether and hydrolysis (0.2N NaOH, MeOH/THF/H_2O) then gave 10, whereas desilylation of 9 generated the second building block (11) (Bu_4NF, THF). Condensation of carboxylic acid (10) and alcohol (11) via the Keck protocol (DCC, DMAP, CSA, CH_2Cl_2) and desilylation of the resultant ester (12) (AcOH/THF/H_2O) produced 13. The third fragment, TBDMS ether (14) of (3S)-hydroxybutyric acid and coupled with 13 (DCC, DMAP, CSA, CH_2Cl_2). Removal of the silyl and t-butyl moieties in 15 (TFA/CH_2Cl_2/thioanisole) provided seco acid (16), which smoothly underwent Yamaguchi macrolactonization (DMAP, 2,4,6-trichlorobenzoyl chloride). Finally, deprotection of 17 (TFA/CH_2Cl_2) gave synthetic 1, identical in all respects with a sample of the natural product. We have studied on the constituents and the stereochemical aspects of lupin alkaloids in differentiated plants and undifferentiated tissue culture of Thermopsis species in Leguminous plants. A new alkaloid, (+)-lupanine N-oxide (14), have been isolated from T. lupinoides with nine known alkaloids (11-15, 17, 18, 21-23) (scheme 2). Two new alkaloids, (-)-O-acetylbabtifoline (19) and (-)-tetrahydrocytisine (24), have been isolated from T. chinensis with nine known alkaloids (11,12,16,17,18,20-23). The structure of new alkaloids were determined by spectroscopic methods and by chemical transformations. We have also studied on the production of alkaloids with callus culture of T. lupinoides and Sophora flavescens. The differentiated plants of T. lupinoides and T. chinensis accumulated (+)-lupanine (11)(7S:9S:11S) series alkaloids together with the antipodal (-)-anagyrine (17)(7R:9R:11R) series. It is interesting from the biosynthetic point of view that the sparteine-type alkaloids (11-14) and the α-pyridone-type alkaloids (16-23) have opposite absolute configuration with each other. However, (+)-lupanine (11)(6R:7S:9S:11S) was the only detectable alkaloid in the green callus of T. lupinoides. In the green callus of S. flavescens, matrine was the alkaloid mainly. The multiple shoots of S. flavescens additionally produced not only matrine but also 5,6-dehydrolupanine (16) and anagyrine (17). These results suggest that the biosynthesis of (+)-lupanine (11) and matrine, both of which are assumed to be the early intermediates in biosynthesis of lupin alkaloids, are related to greening of the tissues in Thermopsis and Sophora (scheme 1)., 天然有機化合物討論会実行委員会 - Glycosidic alkaloids from Lupinus hirsutus

Genetic analysis of production process in secondary metabolites reveals their biosynthetic pathways and their regulatory networks for gene expression. These results reflect on not only basic research but also application to the industrial production process. But almost Streptomyces microorganisms that produce valuable secondary metabolites including antibiotics cannot be applied to molecular genetic approaches (DNA transformation, gene targeting and so on). In this study, we have developed a versatile host for the production of exogenous secondary metabolites using efficient heterologous gene expression system. An industrial anthelmintic antibiotic, avermectin, producer S. avermitilis was used as this model and its chromosome has been engineered for heterologous expression of gene cluster for secondary metabolite biosynthesis. The results indicate that the versatile host constructed is useful for the heterologous expression of many gene clusters for secondary metabolite biosynthesis. - Metabolomics of avermectin-producing strain, Streptomyces avermitilis
