Abstract
Cellular senescence is a complex stress response that results in the permanent arrest of cell proliferation. The accumulation of senescent cells occurs during aging in living organisms, and contributes to tissue dysfunction. Although there are growing lines of evidence that various metabolic changes occur in senescent cells, the link between cellular metabolism and senescence is not yet fully understood. In this study, we demonstrate that alterations in the metabolism of branched-chain amino acids (BCAAs) play a crucial role in establishing cellular senescence. Furthermore, we identified mitochondrial BCAA transamination as a crucial step in this process. Our findings show that various types of cellular stress lead to a reduction in the expression of BCAA aminotransferase 2 (BCAT2), one of the BCAA catabolic enzymes, resulting in decreased catabolism of BCAAs and reduced synthesis of glutamate. The reduction of BCAA catabolites, together with the consequent limitation in glutathione production from glutamate, triggers cellular senescence. Furthermore, we demonstrate that a reduction in BCAT2 levels alone is sufficient to induce cellular senescence, both in cultured cells and in mice. Additionally, our results demonstrate that aging alters BCAA metabolism in both mice and humans. Our findings provide new insights into the metabolic mechanisms underlying cellular senescence, with a particular focus on the role of BCAAs. - GDE5/Gpcpd1 activity determines phosphatidylcholine composition in skeletal muscle and regulates contractile force in mice

Abstract
Glycerophosphocholine (GPC) is an important precursor for intracellular choline supply in phosphatidylcholine (PC) metabolism. GDE5/Gpcpd1 hydrolyzes GPC into choline and glycerol 3-phosphate; this study aimed to elucidate its physiological function in vivo. Heterozygous whole-body GDE5-deficient mice reveal a significant GPC accumulation across tissues, while homozygous whole-body knockout results in embryonic lethality. Skeletal muscle-specific GDE5 deletion (Gde5 skKO) exhibits reduced passive force and improved fatigue resistance in electrically stimulated gastrocnemius muscles in vivo. GDE5 deficiency also results in higher glycolytic metabolites and glycogen levels, and glycerophospholipids alteration, including reduced levels of phospholipids that bind polyunsaturated fatty acids (PUFAs), such as DHA. Interestingly, this PC fatty acid compositional change is similar to that observed in skeletal muscles of denervated and Duchenne muscular dystrophy mouse models. These are accompanied by decrease of GDE5 expression, suggesting a regulatory role of GDE5 activity for glycerophospholipid profiles. Furthermore, a DHA-rich diet enhances contractile force and lowers fatigue resistance, suggesting a functional relationship between PC fatty acid composition and muscle function. Finally, skinned fiber experiments show that GDE5 loss increases the probability of the ryanodine receptor opening and lowers the maximum Ca2+-activated force. Abstract
Silicon nanowire (SiNW) biosensors, operating as field-effect transistors (FETs), demonstrate remarkable capabilities for the ultrasensitive detection of specific biomolecules. Our prior work specifically explored the impact of SiNW widths on biosensor sensitivity, highlighting that narrower SiNWs significantly enhance detection sensitivity. While experimental studies provide valuable insights, theoretical investigations into the combined effect of multiple parameters on sensing performance are crucial. However, theoretical studies have been relatively scarce in the research of SiNW biosensors. In response to this gap, we developed a numerical model of SiNW biosensor using the finite-element method in COMSOL Multiphysics. By leveraging simulations, we explored the sensing performance of SiNW biosensors across various widths, thicknesses, impurity concentrations, and their combined effects, addressing a previously unexplored area in this research. Based on the simulations, the optimal structure that exhibits both high sensitivity and measurable current was predicted. To ascertain the reliability of our simulations, a subset of the results was compared with experimental data. Our findings indicate the potential for achieving ultrasensitive biomolecule detection using SiNW biosensors through structural optimization. - Clinical effect of phytosterol-enriched saw palmetto extract on urinary function in healthy middle-aged and older males: a double-blind, placebo-controlled randomized comparative study

Background: Saw palmetto extract (SPE) is one of the most widely used supplements for benign prostatic hyperplasia (BPH) due to its pharmacological effects. β-sitosterol-enriched saw palmetto oil (VISPO™) contains more than 3% β-sitosterol and 85% total fatty acids and has been shown to improve symptoms in patients with BPH. Lower urinary tract symptoms (LUTS) require primary preventive measures in countries with accelerated population aging, including Japan. VISPO™ is expected to be effective against the early symptoms of LUTS. However, the effect of VISPO™ on urinary function in healthy individuals has not been elucidated.Objective: This clinical study evaluated the efficacy and tolerability of VISPO™ in the management of urinary complaints among healthy middle-aged and older men who did not require treatment.Methods: We conducted a randomized, double-blind, placebo-controlled trial that included 54 male participants who were assigned to consume VISPO™ or placebo (olive oil) capsules for 12 weeks. Biometric data, uroflowmetry results, and salivary hormone levels were assessed at weeks 1 and 12. Urinary function and quality of life were assessed at weeks 1, 4, 8, and 12 using the International Prostate Symptom Score (IPSS), Overactive Bladder Symptom Score (OABSS), Core Lower Urinary Tract Symptom Score (CLSS), and WHOQOL-26.Results: Two-way analysis of variance revealed that IPSS and OABSS were significantly lower in the VISPO™ group that in the placebo group. However, there were no significant differences in urine volume, urinary flow rate, or salivary hormone levels. In addition, no participants reported adverse effects (diarrhea, abdominal pain, etc.). Conclusion: The results of the present study, which is the first randomized controlled trial of VISPO™ conducted among healthy participants, demonstrate the effectiveness of VISPO™ in the management of lower urinary tract complaints that do not require treatment. Silicon nanowire (SiNW) biosensors have attracted a lot of attention due to their superior sensitivity. Recently, the dependence of biomolecule detection sensitivity on the nanowire (NW) width, number, and doping density has been partially investigated. However, the primary reason for achieving ultrahigh sensitivity has not been elucidated thus far. In this study, we designed and fabricated SiNW biosensors with different widths (10.8-155 nm) by integrating a complementary metal-oxide-semiconductor process and electron beam lithography. We aimed to investigate the detection limit of SiNW biosensors and reveal the critical effect of the 10-nm-scaled SiNW width on the detection sensitivity. The sensing performance was evaluated by detecting antiovalbumin immunoglobulin G (IgG) with various concentrations (from 6 aM to 600 nM). The initial thickness of the depletion region of the SiNW and the changes in the depletion region due to biomolecule binding were calculated. The basis of this calculation are the resistance change ratios as functions of IgG concentrations using SiNWs with different widths. The calculation results reveal that the proportion of the depletion region over the entire SiNW channel is the essential reason for high-sensitivity detection. Therefore, this study is crucial for an indepth understanding on how to maximize the sensitivity of SiNW biosensors. - PPARα activation partially drives NAFLD development in liver-specific Hnf4a-null mice

Abstract
HNF4α regulates various genes to maintain liver function. There have been reports linking HNF4α expression to the development of non-alcoholic fatty liver disease (NAFLD) and non-alcoholic steatohepatitis. In this study, liver-specific Hnf4a-deficient mice (Hnf4aΔHep mice) developed hepatosteatosis and liver fibrosis, and they were found to have difficulty utilizing glucose. In Hnf4aΔHep mice, the expression of fatty acid oxidation-related genes, which are PPARα target genes, was increased in contrast to the decreased expression of PPARα, suggesting that Hnf4aΔHep mice take up more lipids in the liver instead of glucose. Furthermore, Hnf4aΔHep/Ppara−/− mice, which are simultaneously deficient in HNF4α and PPARα, showed improved hepatosteatosis and fibrosis. Increased C18:1 and C18:1/C18:0 ratio was observed in the livers of Hnf4aΔHep mice, and the transactivation of PPARα target gene was induced by C18:1. When the C18:1/C18:0 ratio was close to that of Hnf4aΔHep mouse liver, a significant increase in transactivation was observed. In addition, the expression of Pgc1a, a coactivator of PPARs, was increased, suggesting that elevated C18:1 and Pgc1a expression could contribute to PPARα activation in Hnf4aΔHep mice. These insights may contribute to the development of new diagnostic and therapeutic approaches for NAFLD by focusing on the HNF4α and PPARα signaling cascade. - Characterization of recombinant murine GDE4 and GDE7, enzymes producing lysophosphatidic acid and/or cyclic phosphatidic acid.

GDE4 and GDE7 are membrane-bound enzymes that exhibit lysophospholipase D (lysoPLD) activities. We found that GDE7 produced not only lysophosphatidic acid (LPA) but also cyclic phosphatidic acid (cPA) from lysophospholipids by a transphosphatidylation reaction. In contrast, GDE4 produced only LPA. The analysis of substrate specificity showed that 1-alkyl-lysophosphospholipids were preferred substrates for both enzymes rather than 1-alkyl-lysophospholipids and 1-alkenyl-lysophospholipids. Among the various lysophospholipids with different polar head groups that were tested, lysophosphatidylglycerol and lysophosphatidylserine were preferred substrates for GDE4 and GDE7, respectively. The detailed analysis of the dependency of the enzyme activities of GDE4 and GDE7 on divalent cations suggested multiple divalent cations were bound in the active sites of both enzymes. Type 1 diabetes (T1D) is an autoimmune disease in which insulin-producing pancreatic β-cells are destroyed. Intestinal helminths can cause asymptomatic chronic and immunosuppressive infections and suppress disease in rodent models of T1D. However, the underlying regulatory mechanisms for this protection are unclear. Here, we report that CD8+ regulatory T (Treg) cells prevent the onset of streptozotocin -induced diabetes by a rodent intestinal nematode. Trehalose derived from nematodes affects the intestinal microbiota and increases the abundance of Ruminococcus spp., resulting in the induction of CD8+ Treg cells. Furthermore, trehalose has therapeutic effects on both streptozotocin-induced diabetes and in the NOD mouse model of T1D. In addition, compared with healthy volunteers, patients with T1D have fewer CD8+ Treg cells, and the abundance of intestinal Ruminococcus positively correlates with the number of CD8+ Treg cells in humans. - The role of autonomously secreted PGE2 and its autocrine/paracrine effect on bone matrix mineralization at the different stages of differentiating MC3T3-E1 cells.

Sphingosine 1-phosphate (S1P) is a potent lipid mediator that modulates inflammation and angiogenesis. In this study, we investigated the possible involvement of S1P in the pathology of light-induced retinal degeneration in vivo and in vitro. The intracellular S1P and sphingosine kinase (SphK) activity in a photoreceptor cell line (661W cells) was significantly increased by exposure to light. The enhancement of SphK1 expression was dependent on illumination, and all-trans-retinal significantly promoted SphK1 expression. S1P treatment reduced protein kinase B (Akt) phosphorylation and increased the protein expression of cleaved caspase-3, and induced photoreceptor cell apoptosis. In vivo, light exposure enhanced the expression of SphK1 in the outer segments of photoreceptors. Intravitreal injection of a SphK inhibitor significantly suppressed the thinning of the outer nuclear layer and ameliorated the attenuation of the amplitudes of a-waves and b-waves of electroretinograms during light-induced retinal degeneration. These findings imply that light exposure induces the synthesis of S1P in photoreceptors by upregulating SphK1, which is facilitated by all-trans-retinal, causing retinal degeneration. Inhibition of this enhancement may be a therapeutic target of outer retinal degeneration, including age-related macular degeneration. - Regulation of MC3T3-E1 differentiation by actin cytoskeleton through lipid mediators reflecting the cell differentiation stage.

Actin cytoskeleton is reported to be related in various functions of osteoblast, bone-forming cell. However the function of actin cytoskeleton in osteoblasts is not fully understood, since bone formation is derived from extracellular interactions of functional proteins produced from osteoblasts, including osteocalcin (Ocn), and it is a result of closely and complex organized sequence of biochemical events. In this study, we showed that actin cytoskeleton of MC3T3-E1 cells functioned in recognition of cell condition and regulation of extracellular matrix mineralization, bone formation. Maturation of MC3T3-E1 cells by 14 days of culture reduced F-actin filaments, while induced expression of Ocn mRNA known as late stage differentiation marker and matrix mineralization, terminal stage of cell differentiation. The disruption of actin cytoskeleton with Cyto D in immature MC3T3-E1 cells significantly increased expression of Ocn mRNA in 24 h. Both PTX-induced inhibition of signal transduction through GPCRs and celecoxib-induced suppression of lipid mediators in immature MC3T3-E1 cells reduced actin filaments and suppressed matrix mineralization. Furthermore, addition of lipid mediators extracted from culture mediums of differentiated MC3T3-E1 cells by Bligh-Dyer method induced actin cytoskeleton reorganization and matrix mineralization change in MC3T3-E1 cells. Taken together, our data suggest that actin cytoskeleton of MC3T3-E1 cells regulates activation of developmental pathway reflecting cell differentiation stages through lipid mediators. A simple fabrication process of an n-type silicon nanowire (SiNW) biosensor for sub-10 femtomolar (fM) concentration immunoglobulin detection was presented in this work. The SiNWs with different widths of 80-190 nm were fabricated using electron beam lithography and reaction ion etching techniques. The electrical characteristics of SiNWs with various widths were measured. And it can be observed that thin SiNW has high resistance, which is in agreement with electrical resistance theory. Furthermore, the surface of the fabricated SiNW was functionalized by 3-aminopropyltriethoxysilane for making the biosensor device to detect the binding of immunoglobulin G (IgG) molecules. The responsivity of the biosensor was investigated by observing electrical performance in response due to IgG with various concentration from 6 fM to 600 nanomolar (nM). The resistance changing ratio based on the current voltage (I -V ) characteristics was analyzed and it increased with increasing of the IgG concentration. BACKGROUND: Cysteinyl leukotrienes (LTs) are key mediators in inflammation. To explore the structure of the antigen-recognition site of a monoclonal antibody against LTC4 (mAbLTC), we previously isolated full-length cDNAs for heavy and light chains of the antibody and prepared a single-chain antibody comprising variable regions of these two chains (scFvLTC). METHODS: We examined whether mAbLTC and scFvLTC neutralized the biological activities of LTC4 and LTD4 by competing their binding to their receptors. RESULTS: mAbLTC and scFvLTC inhibited their binding of LTC4 or LTD4 to CysLT1 receptor (CysLT1R) and CysLT2 receptor (CysLT2R) overexpressed in Chinese hamster ovary cells. The induction by LTD4 of monocyte chemoattractant protein-1 and interleukin-8 mRNAs in human monocytic leukemia THP-1 cells expressing CysLT1R was dose-dependently suppressed not only by mAbLTC but also by scFvLTC. LTC4- and LTD4-induced aggregation of mouse platelets expressing CysLT2R was dose-dependently suppressed by either mAbLTC or scFvLTC. Administration of mAbLTC reduced pulmonary eosinophil infiltration and goblet cell hyperplasia observed in a murine model of asthma. Furthermore, mAbLTC bound to CysLT2R antagonists but not to CysLT1R antagonists. CONCLUSIONS: These results indicate that mAbLTC and scFvLTC neutralize the biological activities of LTs by competing their binding to CysLT1R and CysLT2R. Furthermore, the binding of cysteinyl LT receptor antagonists to mAbLTC suggests the structural resemblance of the LT-recognition site of the antibody to that of these receptors. Leukotrienes (LTs) are 5-lipoxygenase metabolites derived from arachidonic acid and play important roles in many inflammation and allergic disorders. They are classified to two groups, LTB4 and peptide-LTs. Both groups are thought to show their biological activities through their cellsurface receptors, but no LT receptor had been purified to homogeneity or cDNA-cloned. Recently, we cloned a cDNA for a LTB4 receptor from HL 60 cells. The open reading frame of the cDNA encodes a protein of 352 amino acids and is predicted to contain seven membrane-spanning domains. In CHO cells stably expressing the LTB4 receptor, LTB4 elicited many signal transductions such as increase in intracellular calcium, InsP3 accumulation, and inhibition of adenylyl cyclase. The CHO cells revealed a marked chemotactic response toward nM order of LTB4. Next, we analyzed the signal transduction mechanisms through LTD4 receptors using human monocytic leukemia THP-1 cells. When these cells were stimulated with LTD4, intracellular calcium concentration was increased and MAP kinase was activated severalfold. This activation was mediated a protein kinase Cα-Raf-1 dependent pathway. A chemotactic response of THP-1 cells toward LTD4 was observed. Pulmonary involvement with mucormycosis is quite uncommon, but the incidence has been increasingly recognized, especially in patients with leukemia or lymphoma. We report three autopsied cases of generalized mucormycosis (phycomycosis) in patients with non-Hodgkin's lymphoma, acute exacerbation of chronic myelogenous leukemia, and acute myelogenous leukemia. All patients had abnormal lung shadows and high fever unresponsive to broad-spectrum antibiotics and trimethoprim-sulfamethoxazole.
Their chest X-ray findings were as follows;
Cases 1: Diffuse ground glass appearance with nodular density at the right hulim suggesting hemorrhagic infarction.
Cases 2: Diffuse ground glass appearance.
Cases 3: Several patchy infiltrates in the right lung and a large nodular density with cavities in the left lung field.
In two of three cases, flucytosine was administered without any remarkable effect. Histological examination revealed multiple infarction and thrombosis in various parts of pulmonary arteries in which broad and nonseptate hyphae were found. Sputum cultures were performed in all cases white they were alive, but no pathognomonic organism was found. Premortal diagnosis of mucormycosis is extremely difficult, but one possible way of diagnosis might be by lung perfusion scanning using 131I-MAA. - A Case of Sino-Bronchiectasis Associated with Lack of HLA A, B, and C Antigens.

We present a case of diffuse sino-bronchiectasis associated with the lack of expression of HLA A, B, and C antigens. The case was a 40-year-old male diagnosed in our outpatient clinic as sino-bronchiectasis at about ago 20. In these 20 years, bronchiectasis has progressed gradually. Twenty years ago, bronchiectasis was limited to bilateral middle lobe bronchi, but now it extends to all bronchi.
In this patients, we also found the lack of expression of HLA A, B, and C antigens on his lymphocytes. Concerning other immunological data, his serum IgA, α1-antitrypsin, and β2-microglobulin levels were increased, and serum IgG, IgM and complements levels were within normal limits. Lymphocyte stimulation tests using, PHA, PWM, and Con-A show slightly low but positive reactions. Also, in the analysis of lymphocyte subsets the Leu-3aILeu-2a ratio showed a slight increase. It has been suggested that HLA molecules play an important role in immunological cellular interactions, especially between T-lymphocytes and other cells.
Therefore, we suspect that disorders of surface antigens on the lymphocytes of this case could be closely related to the respiratory tract disorders. - 胸腺原発が疑われた Squamous cell carcinoma の1例

It has been demonstrated that many different bronchoactive agents can cause a vagally mediated bronchoconstriction in animals and in man. Although the effects of local anesthetic aerosols on reflex bronchoconstriction have been investigated in the dog, little is known about the effects of intravenous general anesthetic commonly used in animal experiments on the reflex component of vagus nerve-induced bronchoconstriction.
We anesthetized 32 young mongrel dogs, weighing 6 to 10kg, with chloralose (30mg/kg), paralyzed the respiratory muscle with succinylcholine chloride, and ventilated the lungs through a tracheal cannula at a constant tidal volume (150-250ml) and a frequency of 16/min. Catheters were inserted into the left femoral artery and vein, and measurement of systemic arterial blood pressure (Psyst) and transfusion or infusion of drugs were conducted, respectively. The pressure transducer catheter was connected to the orifice of the tracheal cannula to monitor tracheal pressure (Ptr). The right side vagosympathetic nerve (V.N.) was isolated at the level of thyroid cartilage. We stimulated the cervical V.N. electrically with 5 to 30C from a square-wave stimulator (ME-6052); pulse duration was 1ms with a frequency of 30shocks/s.
1) The percent change of Ptr showed a marked increase by increasing stimulation voltage.
2) The percent increase of Ptr induced by V.N. stimulation showed a dose-dependent decrease with intravenous administration of pentobarbital sodium and ketamine hydrochloride.
3) The percent increase of Ptr suppressed by an intravenous infusion of pentobarbital sodium and ketamine hydrochloride showed a dose-dependent recovery by intravenous administration of neostigmine bromide.
4) The percent increase of Ptr potentiated by neostigmine bromide was suppressed dose-dependently by intravenous administration of pentobarbital sodium.
5) The percent increase of Ptr induced by an intravenous administration of acetylcholine and prostaglandin F2α did not show any change by administration of pentobarbital sodium. - びまん性汎細気管支炎の内視鏡像と中枢気道粘膜組織像

Transbronchial lung biopsy (TBLB) is used for the diagnosis of pulmonary nodules and diffuse lung disease. Reports of complications have been infrequent, but deaths from hemorrhage have been reported. In the present investigation patients who suffered from hemorrhage induced by TBLB, bronchial mucous membrane biopsy and from local injection of anticancer drugs were treated with thrombin solution. Hemorrhage (30-200ml) occurred in 12 of 122 tumor biopsy cases, 14 of 170 TBLB cases, 2 of 102 local anticancer drug injection procedures and 1 of 20 bronchial mucous membrane biopsy cases. For these 29 cases of hemorrhage, we instilled 5 ml of thrombin solution (100 U/ml). In 27 patients, hemorrhage stopped after instillation of one dose of thrombin, and in the 2 other patients stopped after a second dose within 2 minutes. The avove results suggest that thrombin solution instillation therapy might be a useful method to treat hemorrhage induced by the various fiberoptic procedures. - 喀痰よりPasteurella ureaeが検出された慢性気管支炎の1症例

Bronchial hyperreactivity to nonspecific bronchoconstrictor stimuli is one of the diagnostic features of asthma. In the present investigation we attempted to determine whether various bronchoactive agents act on bronchoconstriction induced by vagosympathetic nerve stimulation.

Catheters were inserted into the left femoral artery and vein, and measurement of systemic arterial blood pressure (Psyst) and transfusion or infusion of drugs were conducted, respectively. The pressure transducer catheter was connected to the orifice of the tracheal cannula to monitor tracheal pressure (Ptr). The right side vagosympathetic nerve was isolated at the level of thyroid cartilage. We stimulated the cervical vagosympathetic nerve electrically with 5-25V from a square-wave stimulator (ME-6052); pulse duration was lms and the frequencies were 30shocks/s.
1) Percent change of Ptr showed a maked increase by increasing stimulation voltage and reached a plateau at 15 V.
2) Percent change of Ptr induced by vagosympathetic nerve stimulation showed a marked increase 5min after intravenous infusion of neostigmine bromide, and thereafter it showed a gradual decrease and returned to the control level after 75min.
3) Percent change of Ptr induced by vagosympathetic nerve stimulation showed a significant increase with continuous infusion of PGF2α, serotonin, histamine, Tx B2, LT C4 and LT D4.
4) Percent change of Ptr induced by vagosympathetic nerve stimulation showed a significant decrease with continuous infusion of isoproterenol, PGE2, 6-keto PGF1α and aminophylline.
5) Percent change of Ptr induced by vagosympathetic nerve stimulation ahowed a marked increase after pretreatment with propranolol dose-dependently, and reached a plateau at the dose of 600μg/kg.
6) Percent change of Ptr induced by vagosympathetic nerve stimulation showed a significant increase after pretreatment with propranolol and showed a significant decrease after pretreatment with phenoxybenzamine.
The above results suggest that bronchoconstrictor responses induced by vagosympathetic nerve stimulation were modulated by various vasoactive substances. - 老年者の気管支喘息に対するHC20-511 (ketotifen) の臨床効果

An 18 year-old man was admitted to our hospital complaining of a small amount of bloody sputum and abnormal chest roentgenograms. He had several congenital skin angiomas. The colours of these angiomas were reddish purple, and turned pale on compression. Laboratory examinations showed almost normal values, but blood gas analysis revealed mild hypoxemia. Chest roentgenograms showed mediastinal widening and diffuse reticulo-nodular shadows throughout both lungs. These findings on chest radiograms had been recognized since he was 6years old, but the etiology had not been elucidated. Bronchoscopy demonstrated telangiectasis in bronchial mucosa and soft masses like varices along the left main bronchus. Pulmonary angiograpy showed narrowing of the left main pulmonary artery. Bronchial arteriography revealed elongation and dilatation of both bronchial arteries and early filling of the contrast media into the mediastinal and pulmonary masses. The main draining vessels were pulmonary veins. These findings were compatible with diffuse hemangiomas in the mediastinum and in the lung. - Direct β‐Adrenergic Receptor Assay of Guinea Pig Lung :Effects of α, β-Adrenergic Receptor Blockers

The lung is one of the organs that possess abundant β-adrenergic receptors. Direct radiologic binding methods for studying β-adrenergic receptors have been developed recently. These methods can provide new insights into the molecular characteristics and regulatory properties of β-adrenergic receptors in the lung.
Recently, α-adrenergic receptor blockers have been used for the therapy of brochial asthma and some good theraeputic effects have been reported. However the mechanisms have not been elucidated yet.
β-adrenergic receptor blockers are widely used for patients with hypertension or ischemic heart diseases, and sometimes the use of β-adrenergic receptor blockers elicits asthmatic attacks in patients with bronchial asthma. But actually the incidence of such attacks is not so high as could be expected.
In the present investigation we determined the number of β-adrenergic receptors in the lung membrane of guinea-pigs, weighing 150-280g. by direct methods using [3H]-(-)-dihydroalprenolol ([3H]-DHA). Effects of phentolamine and propranolol on the β-adrenergic receptors in the lung were also investigated.
1) The mean values and standard deviation of the number of β-adrenergic receptors (the concentration of [3H]-DHA binding sites) and the dissociation constant (Kd) were 789±121f mol/mg protein and 1.14±0.22nM, respectively (n=10).
2) The treatment of guinea-pigs with phentolamine (1mg/kg, subcutaneously, for 6 days) induced a significant increase in the number of lung β-adrenergic receptors, 971±149fmol/mg protein (n=8, p<0.02), while Kd did not show any significant change. This increase in the number of lung β-adrenergic receptors might be one of the mechanisms of α- adrenergic receptor blockers in bronchial asthma.
3) The treatment of guinea-pigs with propranolol (1mg/kg, subcutaneously, for 6 days) induced a significant increase in the number of lung β-adrenergic receptors, 931±141fmol/mg protein (n=8, p<0.05), while Kd did not show any significant change. This increase in the number of lung β-adrenergic receptors might be one of the reasons why the incidence of asthmatic attacks is not so high in spite of the widespread use of β-adrenergic receptor blockers. - 頸部・縦隔リンパ節腫脹, 胸・腹水貯留, 皮疹など多彩な症状を呈した結核の1症例

G2A is a G protein-coupled receptor that uses oxidized free fatty acids as ligands. The addition of oxidized free fatty acids such as 9-HODE to the cultured cells resulted in cell cycle arrest. G2A induction was also observed by stress stimulation such as glucose starvation and hypoxia. It became clear that G2A induction is deeply involved in changes in intracellular pH and glutathione concentration. When glioma cells expressing G2A were transplanted into immunodeficient mice, tumor-formation and metastasis to the organs was observed. Immunohistochemical analysis of metastatic lesion revealed that epithelial mesenchymal transition occurred.
Thus, G2A, a stress-inducible GPCR, stops the cell cycle in response to oxidized free fatty acids. On the other hand, G2A increases stemness in tumor cells resulting in epithelial mesenchymal transition and forms metastatic foci. - Analysis of receptor signaling in living cells by surface plasmon resonance

Various methods have been used for the analysis of intracellular signaling from cell membrane receptors. However, there are problems in the reproducibility and operability. Previously, we have succeeded in measuring the SPR (surface plasmon resonance) through GPCR (G-protein coupled receptor) in living cells. In this study, we elucidated the part of the intracellular signaling pathways of GPCR signal using various inhibitors. Furthermore, we succeeded in measuring the SPR though RTKs (tyrosine kinase receptors), another group of membrane receptors. A purpose of this study was to clarify the physiological and pathological functions of a G-protein coupled receptor called G2A, and was to investigate the basic aspects of G2A signaling to establish new diagnosis / therapy. G2A is a receptor which ligands are oxidative flee fatty acids such as 9-HODE and 11-HETE derived from linoleic acid or arachidonic acid, respectively. It was reported that G2Awas induced by various DNA damage stimulation and its stimulation causes arrest of cell cycle. However, the physiological function of G2Awas unidentified.
As skin is routinely and pathologically exposed to many oxidative stresses such as UV radiation, chemical agents, and inflammation that might induce both G2A expression and production of G2A ligands, we examined G2A fraction in human keratinocytes. G2A was expressed in human epidermis and normal human epidermal keratinocytes (NHEK). 9-HODE evoked intracellular calcium mobilization, and secretion of cytokines including IL-6, IL-8, and GM-CSF in NHEK cells. These responses became prominent in HaCaT cells, an immortalized human keratinocyte cell line, by overexpression of G2A. 9-HODE further inhibited proliferation of NHEK cells by suppressing DNA synthesis and arresting the cell cycle in the G0/1-phase, and down-regulation of G2A expression by small interfering RNA caused suppression of 9-HODE-induced effects on proliferation of NHEK cells 13-HODE, much weaker ligand of G2A, did not show any effects on either cytokine secretion and proliferation in NHEK cells. On the other hand, UVB and hydrogen peroxide induced G2A expression and caused oxidation of linoleate to produce 9-HODE in HaCaT cells.
These results suggest that 9-HODE-G2A signaling plays proinflammatory roles in skin under oxidative conditions. - 生体情報の受容伝達と機能発現

Upon cell stimulation, various types of phospholipases are activated, and a class of lipid mediators are produced. They play important role in self-defense and maintenance of homeostasis. In the last three years, we have carried out biochemical and genetic experiments, and obtained results as follows. The results have been published in almost 50 original articles and 6 reviews or book chapters.
1. Studies on cytosolic phospholipase A2 and related molecules.
cPLA2α-null mice showed impairment in synaptic plasticity and reproduction. The enzyme also plays important roles in inflammatory bone resorption, autoimmune arthritis, acute respiratory distress syndrome (ARDS) and bleomycin-induce pulmonary fibrosis. Thus, a selective cPLA2a inhibitor would be beneficial for these disorders. In addition to a subtype of cPLA2, we also found β, and γ subtypes of the enzyme. While β-subtype has Ca-sensitive C2 domain, g-subtype is activated by reactive oxygen species. We also found a novel truncated isoforms of cPLA2, specifically expressed in dentate gyrus by stimulation.
2. Characterization of leukotriene receptors.
We identified 4 different types of leukotriene receptors (BLT1, BLT2, CysLT1, and CysLT2). They are differently expressed in cells and tissues, by cognate promoter activations. We elucidated detailed signal transduction of these receptors, and established cell, either overexpressing or lacking these receptors. More systematic phenotype analyzes will be done by gene targeting of the mice.
3. Characterization of other types of receptors.
We analyzed PAF (platelet-activating factor) receptor, and further found that PAF is deeply involved in inflammatory responses, capacitation of sperm, and neuronal migration in cerebellar granular cells. We also identified the fate of PAF (its breakdown to lysophospholipids, and reacylation to 1-alky1-phospholipids). 1. Determination of optimal condition of labeling LTB_4 receptor-expressed CHO cells with fluorescent probes.
CHO cells could be adequately labeled for visualization with fluorescent microscopy with 4 micro molar CMFDA or 2 micro molar CMTMR.LTB_4 receptor-expressed CHO cells labeled with this conditions showed the same chemotaxis against LTB_4 as unlabeled cells in chemotaxis assay using chemotaxis chamber.
2. Evaluation of neutrophil accumulation in mice lung with intratracheal administration of various agents and its reproducibility.
As models of neutrophil accumulation in mice lung, we administered LTB_4, bleomycin, and silica intratracheally. After 4 hours of LTB_4 administration, neutrophilia over 10% in bronchoalveolar lavage fluid (BALF) cells were observed in only about 20% of examined mice, and after 24 hours of bleomycin administration, neutrophilia over 50% were observed in only about 50% of examined mice, suggesting that the reproducibility of neutrophil accumulation in mice lung by these methods was not adequate. In case of administration of silica, when silica was recovered in BALF, there were consistent marked neutrophilia (40-80%) in BALF cells 24 hours after silica administration, but, when silica was not recovered in BALF, neutrophilia were not observed. This findings indicated that expectoration and inaccessibility to lung parenchyme of intratracheally administered agents were the main cause of inadequate reproducibility of the procedures. Silica was a suitable agent for intratracheal administration in that its deposition into lung parenchyme could be verified by BALF analysis.
3. in vivo Chemotaxis assay in mice model of intratracheal administration of silica using CMFDA-labeled LTB_4 receptor-expressed CHO cells.
CMFDA-labeled LTB_4 receptor-expressed CHO cells and CMTMR-labeled control CHO cells were injected to mice intravenously via tail veins after 20 hours of intratracheal administration of silica. However, in BALF obtained 4 hours later, neither CMTMR nor CMFDA-labeled cells could not be recovered. Because there was a possibility that LTB_4 receptor-expressed CHO cells was not present freely in alveolar space but attached to pulmonary capillary endothelium or alveolar epithelium, we made histological specimens of lung tissues and examined fluorescent probe-labeled cells, but, at present, we could not obtain any definite results. - INVOLVEMENT OF LIPID MEDIATORS IN INFLAMMATORY DISORDERS

Lipid mediators such as leukotrienes (LTs) and platelet-activating factor (PAF) are major chemical mediators involved in inflammatory and allergic disorders. To clarify the pathological roles of these mediators in inflammatory disorders, this research project was carried out, and following results were obtained.
1. Establishment and analysis of inflammatory animal model
In PAF receptor transgenic mice, PAF receptor deficiency mice, cPLA2 deficiency mice, and wild-type mice, ARDS (adult respiratory distress syndrome) models were made by intratracheal administration of acid or intravenous administration of LPS, and analyzed. The obtained results indicate that PAP and cPLA2-dependent mediators play important roles in these conditions.
PAF receptors are overexpressed in Kupffer cells after experimental hepatectomy in rat.
2. Study on LTB4 receptor
The mechanism of leukocyte-specific expression of LTB4 receptor was revealed by its promoter analysis. Using in vivo chemotaxis of CHO cells expressing LTB4 receptor, the possible roles of LTB4 in pathogenesis of acute renal inflammation were indicated. The second receptor for LTB4 was discovered. The project on establishment of LTB4 deficiency mice is now in progress.
3. Signal transduction through LTD4 receptor
Intracellular signal transduction through LTD4 receptor was analyzed in monocytes and macrophages.
4. Intracellular localization of enzymes involved in production of lipid mediators
The mechanisms of intracellular localization and translocation of enzymes such as cPLA2, 5-lipoxygenase and LTA4 hydrolase were studied.
These results revealed that mechanisms of lipid mediator production and pathological roles of lipid mediators in inflammatory disorders, and may help us develop new therapies and drugs. - FUNCTIONAL REGULATION OF RECEPTORS FOR LIPID MEDIATORS

Lipid mediators such as leukotrienes (LTs) and platelet-activating factor (PAF) are major chemical mediators involved in inflammatory and allergic disorders. Despite their potent biological activities, little is known about the function and translational regulation of their receptors. To clarify these questions, we carried out following experiments and obtained some results that may contribute better understanding of patho-physiological roles of lipid mediators and development of new therapies and drugs for inflammatory and at11ergic disorders.
1. We analyzed the mechanisms of ligand-induced internalization of PAP receptors.
2. Using PAP receptor knockout mice and cPLA2 knockout mice, the roles of lipid mediators in acute lung injuries occurred by LPS and acid were investigated.
3. Functional coupling of LTB4 receptor with G-proteins in porcine WBC was identified.
4. Analysis of human genome for LTB4 receptor was carried out. The mechanisms for leukocyte specific expression of LTB4 receptor was clarified.
5. cDNA clones for LTB4 receptors of mouse, rat and Guinea pig were identified. And their functions were analyzed.
6. We analyzed the signal transduction mechanisms through LTD4 receptors using human monocytic leukemia THP-1 cells. - MECHANISM OF NEURONAL DEATH AND REGENERATION

Neuronal survival and death are determined by multiple factors including neuronal activity as well as an influence from surrounding glia. In this project, we focus on various lipid mediators in terms of its ability of interaction between neuron and glia. We found that various lipid mediators are produced by stimuli, and play roles in the network as follows. LPA, produced by various cells, stimulates growth and cell maturation of neuron as well as proliferation of astrocytes. LPA also releases various cytokines and neurotrophis (NGF, BDNF) from astroglia, which activates neuronal cells. PAP is released from neuron by the NMDA stimulation, and acts on microglia. Microglia produces NO, and causes chemotaxis towards neuron, suggesting the role of microglia for repairing neuronal cells. PAF receptor-deficient mice and phospholipase A2 (cPLA2)-deficient mice are established, and their congenic mice are now available by backcrossing. The exact roles of these lipid mediators will be clarified by extensive analyses of their phenotypes. - IN VIVO AND INTRACELLULAR ROLES OF LEUKOTRIENE B4 RECEPTOR.

To investigate the biological functions of leukotriene B4 receptor in vivo, we established a line of mice deficient in high-affinity leukotriene B4 receptor (BLT1). Embryonic stem (ES) cells were transfected with a targeting vector for BLT1, and several clones were isolated and confirmed for their homologous recombination by Southern blotting. These ES cells were microinjected to blastcysts, transferred to pseudopregnant female mice, and subsequently chimeric mice were obtained. These chimeric mice are crossed among them, and finally BLT1-deficient mice were born. Now these mice are under backcrossing to examine their phenotype.
CHO cells stably expressing BLT1 showed robust chemotaxis toward LTB4 in vitro, so we tried to examine the chemotactic activities of these cells in vivo. Rat acute ischemic-reperfusion renal injury was initiated by 45 minute-clamping of the renal artery. After 24 hrs, cells were injected into the injured kidney, and were allowed to chemotax for 4 hrs. Only in case of ischemic kidney, not in the sham-operated kdney, CHO cells expressing BLT1 migrated into the peritubal portion of the kidney. Control cells transfected with empty vector did not migrate at all. Prefeeding of the rats with BLT antagonist (Ono 4057) not only inhibited the cells migration, but also reduced the severity of the renal failure, suggesting that LTB4-BLT1 interaction plays important roles in the initiation and the progression of ischemic renal failure.(Noiri, E., et al Proc.Natl.Acad.Sci.U.S.A., 2000)
To clarify the molecular mechanisms of the cell-specific expression of BLT1, we examined the transcriptional regulation of BLT1. BLT1 gene is a small gene within 4 kbps, and its promoter contains no TATA-box, but an Initiator sequence. Serial deletion analyses revealed that Sp-1 at-50 from the initiation site plays a pivotal role in BLT1 transcription. Moreover, the promoter region was totally methylated in BLT1-nonexpressing cells, but not in the expressing cells. In vitro methylation of BLT1 promoter almost completely inhibited its promoter activity.(Kato, K., et al.J.Exp.Med.2000)
On the course of analyzing BLT1 promoter, we found a novel gene with structural similarities with BLT1. The encoded protein was a novel G-protein-coupled receptor, and exhibited a low-affinity, but significant binding to LTB4. Leukotrienes are major lipid mediators involved in inflammatory and allergic disorders. Despite their potent biological activities, little is known about the receptors and intracellular signaling pathways.
1.We cloned a cDNA for a LTB_4 receptor (BLT) from HL-60 cells differentiated with retinoic acid using a subtraction strategy. The open reading frame (ORF) of the cDNA encodes a protein of 352 amino acids and is predicted to contain seven membrane-spanning domains. In CHO cells stably expressing BLT, LTB_4 elicited many signal transductions such as increase in intracellular calcium, InsP_3 accumulation, and inhibition of adenylyl cyclase. The CHO cells revealed a marked chemotactic response toward nM order of LTB_4. Calcium increase and InsP_3 accumulation induced by LTB_4 were partially sensitive to pertussis toxin (PTX), while chemotaxis and inhibition of adenylate cyclase were completely sensitive to PTX, indicating that BLT transduces LTB_4 signals through both PTX-sensitive and -insensitive G proteins.
2.We analyzed the signal transduction mechanisms through LTD_4 receptors using human monocytic leukemia THP-1 cells. When these cells were stimulated with LTD_4, intracellular calcium concentration was increased and mitogen-activated protein kinase (MAP kinase) was activated. A chemotactic response of THP-1 cells toward LTD_4 was also observed. We found that LTD_4 has at least two distinct signaling pathways in THP-1 cells, a PTX-insensitive mitogen-activated protein kinase activation through protein kinase Calpha and Raf-1 and a PTX-sensitive chemotactic response. This cellular signaling can explain in part the versatile activities of LTD_4 in macrophages under inflammatory and allergic conditions. - 血小板活性化因子(PAF)受容体の機能解析と制御

Platelet-activating factor (PAF) is a potent phospholipid mediator involved in inflammatory and allergic responses. It is also reported that PAF plays major roles in endotoxin-induced lethality, reproduction and brain development. To determine the role of PAF and its receptor in vivo, we generated a PAF receptor-deficient mice, by honologous recombination. Offspring were obtained with a Menderian distribution. They grow up normally, and no developmental changes are observed in any organs examined. The level of blood immunoglobulins and differential cell counts show no abnormality.
When the mice were sensitized and challenged with ovalbumin, the wild type mice showed a marked bronchial constriction, pulmonary edema, hypotension and death. On the other hand, the PAF receptor-deficient mice showed only moderate response to these treatments. Upon challenging with endotoxin, no significant difference was observed in both groups. Thus, we concluded that PAF is deeply involved in anaphylactic responses, while it shows minor role in the endotoxin-induced pathologies and lethality. Considering PAF antagonists show some protective effects on endotoxin-shock, even in PAF receptor knock-out mice, there may exist other targets of PAF receptor antagonists to protect the living system from endotoxin-induced disorders. - PAF受容体トランスジェニックマウスを用いた気管支喘息発症メカニズムの解明

Endotoxin causes systemic collapse, disseminated intravascular coagulation and lethality, by infection of gram-negative bacteria. Although many reports the production of cytokines, the precise mechanism of pathophysiological events remained obscure. By cell biology and molecular biology techniques, we obtained following results.
(1) Elucidation of relation between endotoxin and PAF receptor
We established transgenic mice overxpressing PAF receptor. These mice showed severe symptoms of endotoxin shock and death by intravenous administration of small doses of LPS.The lethality was efficiently blocked by the use of PAF receptor antagonists, suggesting that PAF-PAFR is involved in the pathogenesis of endotoxin-induced lethality.
(2) Elucidation of LPS action on macrophages
By incubating of macrophages with bacterial endotoxin, PAFR mRNA was increased several fold after 4h. MAP kinase was also activated, and the activation related to the tyrosine phosphorylation of Raf-1 kinase. - トランスジェニックマウスを用いた心筋細胞におけるPAF受容体の解析

Following results were obtained on the physiological role and cellular signaling of platelet-activating factor (PAF).
(1)Cellular signaling via PAF receptor
By using transfected receptor in CHO cells, cellular signaling after PAF application was determined. PAF stimulates phosphoinositide turnover, mitogen-activated protein (MAP) kinase, phospholipase A2, and inhibits adenylate cyclase. From the results of PTX-treatment, PAF receptor appears to couple with two types of G-proteins. Ca-independent activation of MAP kinase was inhibited by wortmannin, suggesting a wortmannin-sensitive molecule such as phosphatidylinositol 3-kinase is present between Gi and MAP kinase.
To determine the molecular mechanisms of signal desensitization by PAF,we prepared CHO cells carrying wild-type, C-terminal deletion mutant, and substitution mutant (Ser, Thr to Ala) receptors. We found that C-terminal cytoplasmic tail is not required for forward signal transduction, but it is essential for signal-shut down. A family of GRK (G-protein receptor kinase) may be involved in the phosphorylation of Ser/Thr residues leading to signal shut down.
(2)Genomic structure of PAF receptor.
Human PAF receptor gene is present as a single copy in haploid on the chromosome 1q. The gene consists of three exons, separated each other > 20kb intron. Two5'-noncoding exons has distinct transcriptional start site, and promoters. These exons are alternatively spliced to a common splice acceptor to yield two different mRNAs. Transcript 1 is present in almost all tissues, and up-regulated by PAF or endotoxin through its NF-kB site. Transcript 2 is expressed in more tissue-specific way, and up-regulated by estrogen, and down-regulated by TGF-b. The physiological and pathological significance of these promoters are currently under investigation. - THE MECHANISM AND BIOSYNTHESIS OF BIOACTIVE LIPIDS

1. STUDY FOR LEUKOTRIENE BIOSYNTHESIS AND METABOLISM
A monoclonal antibody to LTC_4 synthase was developed, and using this antibody enzyme purification is now going on.
A mutagenesis study of LTA_4 hydrolase revealed that the active center for LTB_4 producing activity is different from that for aminopeptidase activity.
We showed that LTB_4 is inactivated by LTB_4-12-hydroxydehydrogenase in porcine kidney, and this enzyme was purified to apparent homogeneity.
2. STUDY FOR FUNCTION OF LEUKOTRIENE
Studies for cDNA cloning of leukotriene receptors are going on using frog oocyte expression system or mammalian cell expression system.
3. STUDY FOR PAR RECEPTOR
The presence of functionally active PAF receptor in rat brain have been indicated.
In a human eosinophilic leukemia cell line (EoL-1 cells), PAF receptor were induced during differentiation by sodium butyrate.
Studies on human PAF receptor gene revealed that two different promoter exist to produce two types of mRNA, namely leukocyte type and heart type.
In CHO cells transfected with PAF receptor, PAF activated MAP kinase and MAP kinase kinase. - Screening and Development of New Immnomodulatory Drugs using PAF Receptor Gene as a Probe

Platelet-activating factor (PAF) is a potent mediator of inflammation, immunological disorders, and endotoxin shock. We have recently isolated cDNA clones of human and guinea pig PAF receptors. Using these clones as probe, we have obtained following results.
1. Isolation and structure of PAF receptor gene.
PAF receptor gene is consisted of three exons which are separated by >20kb introns. All exons are localized on chromosome 1. By alternative splicing, the leukocyte- and heart-type transcripts are produced. They have differentially regulated by distinct promoters.
2. Elucidation of signal transduction through a cloned PAF receptor.
A stable transfectant cell (CHO) was established carrying a cloned PAF receptor. By the application of PAF, phospholipase C and A_2, and MAP (mitogen-associated protein) are all activated, while adenylate cyclase was inhibited. These cellular events appear to be regulated by different types of G-proteins. Furthermore, we found that endotoxin. LPS, stimulated PAF receptor and yielded a Ca increase in human platelets as well as CHO transfectants. Thus, a part of LPS signaling, especially that related to platelet-aggregation might be due to the direct activation of PAF receptor.
3. Development of Screening System of PAF Receptor Agonists and Antagonists.
PAF receptor was expressed in Xenopus oocytes and a sensitive assay system of PAF receptor agonists/antagonists was made. Furthermore, COS cells/CHO cells carrying an excess amount of PAF receptors were constructed. By using membrane fractions of these cells, the highly specific screening system of PAF receptor modulator (either receptor antagonists or uncoupler of receptor-G-protein) was established. Through screening of naturally-occurring and synthetic compounds, several candidate chemicals have been obtained, which have potentiality for immunomodulatory drugs. - Studies on Leukotriene Biosynthesis and Signal Transduction.

PAF and leukotrienes are lipid mediators involved in inflammatory/allergic response as well as various cardiovascular and immunological disorders. We have carried out experiments on biosyntheses and signal transduction of these lipid mediators.
1. Isolation of PAF and leukotriene receptors.
By expression cloning using Xenopus oocyte system, we isolated a cDNA of PAF receptor as the first successful example of receptor cloning of lipid mediators. A human homologue was also obtained. They are composed of 342 amino acid residues with putative seven transmembrane spanning domains. These receptors were expressed in COS cells or CHO cells, and cellular signalings are extensively studied. It was shown that PAF receptor couples to phospholipases, adenylate cyclase, and MAP kinase via different types of G- proteins. Leukotriene receptors (B4, and D4) were solubilized from guinea pig leukocyte and lung membrane, respectively in an active form. They activate phospholipase A_2 and C via Gi proteins. Molecular cloning of these receptors by two strategies (protein purification and expression cloning) is currently ongoing.
2. Leukotriene A_4 hydrolase. a bifunctional enzyme.
Leukotriene B_4, a potent chemotactic factor, is produced by an enzyme termed leukotriene A4 hydrolase. By cDNA cloning and elucidation of the primary structure of the enzyme have shown that the enzyme is highly homologous to various types of metalloproteinases. In fact, it was shown that the enzyme contains a zinc ion, which is essential for both intrinsic aminopeptidase activity and leukotriene A_4 hydrolase. To determine the active site of the enzyme, site-directed mutagenesis experiments were performed. The mutation of His-296, or Glu-315 yielded loss of zinc ion with concomitant disappearance of both enzyme activities. However, by changing Glu-297 to other amino acids. two enzyme activities were clearly separated. The mechanism of incorporation of water molecule as well as the physiological significance of bifunctional role of enzyme remain unclear.
