    Objective:

    This study aimed to determine whether the apparent diffusion coefficients (ADCs) determined by diffusion-weighted imaging (DWI) of magnetic resonance imaging (MRI) could facilitate the malignancy grading of various gliomas.

    Methods:

    Sixty patients with a primary cerebral glioma underwent diffusion-weighted and gadolinium-enhanced (Gd) T1-weighted MRI using a 1.5-T MRI scanner. Scoring was performed based on signal intensities on DWI and Gd images. The mean and minimum ADC values were calculated, and Ki-67 staining was performed for each histological sample to evaluate their tumor proliferative potential. Then, the DWI, Gd, and combined scores were analyzed and compared with the Ki-67 staining index and malignant grade. The relationships among the mean and minimum ADC values, Ki-67 staining index, and malignant grade were also evaluated.

    Results:

    The minimum ADC was inversely correlated with the Ki-67 staining index, with a low minimum ADC suggestive of tumor malignancy. The qualitative evaluation of the D score of water molecule diffusion on DWI accurately reflected the pathological grades of gliomas, with an effectiveness that was at least as good as the quantitative analysis using the minimum ADC. The diagnostic value of Gd images in determining glioma malignancy grades was inferior to that of DWI.

    Conclusion:

    Both DWI and gadolinium-enhanced images of MRI should be considered essential for the diagnosis of tumor malignancy.

    Sonodynamic therapy (SDT) has shown great potential as an approach for cancer treatment, and hyperthermotherapy (HT) is also a promising cancer therapy. Here, we investigate whether HT could improve the efficacy of SDT and to make a preliminary exploration on potential mechanism. Xenograft tumor was established in nude mice model, and SNB19 and U87MG glioma cell lines were utilized for in vitro experiment. Alamar blue assay was performed to assess cell viability. Optical microscope was used to characterize the morphology changes of the glioma cells induced by SDT and HT treatments. Apoptotic rate, mitochondrial membrane potential (MMP), and intracellular production of reactive oxygen species (ROS) were examined by flow cytometer. The cell apoptosis of tumor tissues were detected by TUNEL assay. Furthermore, the expression of apoptosis-related proteins was detected with Western blot in vitro and immunohistochemistry in vivo. SDT plus HT group could significantly reduce the cell viability with circular-cell morphological change, compared with SDT group, and cell viability was decreased depending on raise of 5-ALA concentration, ultrasound exposure time, and temperature. The results also indicate that HT increased a conspicuous apoptosis, ROS production, and a remarkable loss in MMP induced by 5-ALA-SDT in vitro. Meanwhile, our data also demonstrated that the combined treatment could significantly induce apoptosis and delay tumor growth in vivo. Furthermore, in both in vitro and in vivo experiments, SDT plus HT group expressed significantly higher protein levels of Bax and cleaved caspase-3, 8, and 9 compared to SDT, HT, and control groups and significantly lower protein level of bcl-2 than the other three groups, while the expression of these proteins was unchanged between HT and control groups. HT may provide an important promotion on 5-ALA-SDT and further propose that SDT in combination with HT is a new potential application for the treatment of human glioma.

    Bromodeoxyuridine (BrdU) is known to cause base mispairs and DNA strand breaks during DNA synthesis, culminating in growth suppression. To know whether P53 gene plays any role in the BrdU-induced growth suppression, we continuously gave BrdU (20 μM) to the synchronized culture of human gastric cancer cell lines, MKN-45 (P53-wild type) and MKN-28 (P53-mutant), shortly after release from the G1/S block by hydroxyurea. In comparison with the control culture, the growth of MKN-28 was not suppressed until 48 hr of BrdU exposure, while that of MKN-45 was already suppressed at the 24 hr point. Continuous exposure to BrdU caused a S-phase delay and G2 arrest of around 6 hr each in MKN-45 and a delay only of the second S phase in MKN-28 in comparison with synchronized control cultures of matched cell cycle phase. BrdU-exposed MKN-45 cells showed a significantly higher incidence of apoptosis after the cells passed through the first G2 phase and the second S/G2 phases, but MKN-28 did not. It thus appears that the delays of S/G2 phases and an increased incidence of apoptosis in the first cell cycle are p53-dependent. The growth suppression in the second S/G2 phase or later observed in both of the cell lines may be p53-independent.
